Single-molecule spectroscopy and microscopy

چکیده
Advances in detector sensitivity and improvements in instrument design have recently provided scientists with tools to probe single molecules with light, and monitor their photophysical properties with exquisite sensitivity and spatial as well as temporal resolution. Spectroscopic and temporal information is used to explore molecular structure, conformational dynamics, local environment and intermolecular interactions of individual species. High-resolution single-molecule microscopy allows these methods to be used for in vitro or in vivo molecular colocalization with nanometer precision. The collected data have generated a wealth of new information in domains ranging from chemistry and biology to solid state physics. Pour citer cet article : X. Michalet, S. Weiss, C. R. متن کاملMethods of single-molecule fluorescence spectroscopy and microscopy
Optical spectroscopy at the ultimate limit of a single molecule has grown over the past dozen years into a powerful technique for exploring the individual nanoscale behavior of molecules in complex local environments. Observing a single molecule removes the usual ensemble average, allowing the exploration of hidden heterogeneity in complex condensed phases as well as direct observation of dynam...
متن کاملRevisiting atomic force microscopy force spectroscopy sensitivity for single molecule studies
Recently, the rapid advances in quantitative biology and polymer science have led to the atomic force microscope AFM being extensively employed for single-molecule force spectroscopy. Deflection sensitivity, a critical factor in single molecule force spectroscopy, is changed due to the change in bending shape of AFM cantilever when a single molecule is attached to the AFM cantilever tip. We qua...
متن کاملSingle-Molecule Spectroscopy and Dynamics at Room Temperature
Chemists are used to visualizing molecular properties and chemical changes on a single-molecule basis. Holding a molecular model, we can predict the forces with surrounding molecules and conceive a chemical reaction. In contrast, information regarding molecular interactions and chemical dynamics has been primarily derived from experiments conducted on large ensembles of molecules. Although ense...
متن کاملResults and Thoughts on Optical Microscopy Using a Single-molecule Probe
We discuss our recent results as well as planned experiments for obtaining molecular resolution in optical microscopy. By combining single molecule spectroscopy and scanning probe microscopy we have demonstrated optical microscopy with a single-molecule light source. We also sketch a method for the use of an inhomogeneous static electric field for determining the position of a molecule in all t...
متن کاملSingle-molecule fluorescence spectroscopy in (bio)catalysis.
The ever-improving time and space resolution and molecular detection sensitivity of fluorescence microscopy offer unique opportunities to deepen our insights into the function of chemical and biological catalysts. Because single-molecule microscopy allows for counting the turnover events one by one, one can map the distribution of the catalytic activities of different sites in solid heterogeneo...
